I'm setting the startimage as: and I'm trying to use one of the existing folders of the gallery from where the images would be displayed as startup slideshow.

I have 2 questions:

1. Is there any way to exclude thumbnails? (all my images are already of 640x360 in size)

2. Is there any way to make the image selection on random basis? In show.ini, I have already set the imagesorting = rnd, but that didn't seem to work.

Sorry to say the integrated startimage slideshow is not based on the full slideshow moddule, so its a little limited. I dont think you can choose the imagesorting method, and since it simply reads content of a folder, I doubt you can evade thumbnails also. Best would be to create a unique folder -
